Hi,
I was wondering if anyone has experience collecting clinical samples
in the presences of THU, tetrahydrouridine, a cytidine-deaminase
inhibitor.
Are there vendors that supply blood collection tubes containing THU?
Are there vendors that make custom blood collection tubes?
Has anyone had to add THU to blood collection tubes prior to blood
collection at a clinical site?
If you have had experience with any of the above, please reply.

Hi Tara,
I am currently working on a trial that needs collection of blood
samples in THU treated vacutainers. The best way to avoid deamination
is to either collect blood in vacutainers preloaded with 0.05-0.1 ml
of tetrahydrouridine (10mg/ml) or add THU immediately after collection.
I am not sure of any vendors that supply THU coated tubes.

Hi TK,
yes we've had experience with that. I do not know of any vendors for
THU-spiked tubes (and I don't think they exist). What we did:
The THU vial comes in 10 mg. add 1 mL of water (=10 mg/mL)
per 1 mL of blood to be collected you need 10 uL of the THU solution.
take a syring-needle as small as possible (I think they're called
insuline/tuberculine syringes?), anyway, the 0.5 mL type with the
needle attached.
fill the syringe with the amount of THU solution you need (50 uL for
a 5 mL blood tube)
puncture the seal of the vacutainer and add the solution (will be
sucked out of the syringe)
The seal will hold the remaining vacuum, you can even do this a
couple of days in advance.
Do take care to only fill the syringe for the volume needed for one
tube each time, the vacuum will suck out all the contents out of the
syringe.
