Hi,
I would like to ask several questions about Caco-2 permeability and
uptake assay.
1. How to measure TEER? I found when I use the probe to test TEER, the
probe itself could impair the integrity of cell monolayer. Is it OK if
I just put one end of the probe in the basolateral medium, and the
other end in the apical medium without touching the monolayer?
2. How much is normally good TEER value for the differentiated
Caco-2 monolayer? Is 300ohm.cm^2 OK?
3. As for Caco-2 uptake assay, I use it to test the absorption of
highly lipophilic compound as the alternative of permeability assay.
But I found the amount of uptake is quite low. If it is used to
evaluate the endocytosis of nanoparticles, I found the endocytosis is
extremely in the 16~21-day differentiated cells. Is it common
phenomenon?
