We are seeking expert opinion for conducting the preclinical study of
Glimepiride in rats. The highest human dose is 8mg per day.
Glimepiride in combination with Thiazolidone derivative is maximum
dose upto 4mg. The Glimepiride is having linear response range from
0.8 to 30 mg/kg (published in one of the scientific discussion of EMEA
guideline for EU). Toxic dose for Glimepiride is 10g/kg. The technique
use for estimation of drug is HPLC-UV. As to carry out the drug-drug
interaction in rats as with thiazolidone derivative what dose I can
select for present study? We have to convert the human dose to rat
dose or we can take 10mg/kg of drug dose directly. As we want to study
only pharmacokinetic drug-drug interaction. If we are converting the
human dose to rat dose it may not be possible to quantitate drug. The
main objective of the work is to study the drug-drug interactions in
rat model.
