Does anyone know of any instances, preferably documented in the
literature, where low plasma concentrations of an orally administered
drug correlate with systemic AEs (i.e., not GI related AEs)? In this
scenario, the drug in question is not a prodrug and the activities of
the metabolites are unknown. My first suspicion - rapid formation of
offending metabolite(s). Any thoughts?
Thanks,
Burgess

The poster child for this that I can recall is amonifide. See Mark
