dear all now i am going to do method validation for Nicotinine and
cotinine in human plasma, in plasma itself contains some amount of
nicotine and cotinine,so what i will do for this situation, can i
minus the blank response during calculation of CCs & QC? or any other
method is there for this type of the validation,please give me
suggestion.

hi gopinath,
i had faced this problem when i had chosen caffeine as an internal
standard. You can minus the blank response and proceed for validation
or i would suggest that you optimize your extraction methodology by
treating the plasma to obtain a clear blank. Try LLE

If you have a pre-dose sample(s) you can determine the levels for each
patient and subtract this value from the post dose values.
