Dear all
I am a PhD student of Pharmaceutical Science. And now I want to do
some experiment to confirm Pharmacodynamics of 001 (compound number).
This compound has anti-inflammation activity. Because it is a little
hard to determine the blood concentration of compound 001 under
regular dosing, we want to get more information for dose regimen
optimization by pharmacodynamic research.
My idea is as below. Welcome any comments in order to give a
scientific experiment design:
(1). First, we want to establish an inflammation model with injection
of LPS, and then detect these pro-inflammatory cytokines in blood
sample at different time points. For example TNF-, so we could know
after injection of LPS, when TNF- will give its peak level expression.
The time will be regarded as Tmax (TNF-).
(2)Second, we want to treat the animal with compound 001 at different
time points before injections of LPS, for
example,-24h,-16h,-12h,-8h,-4h, and 0h. And then injection of LPS,
flowing detecting the concentration of TNF- at the expected Tmax
(TNF-). This aimed to see after how long time the compound will have
no affect on LPS induced inflammation model.
(3)I still want to whether the surgery for collecting blood sample
will have some infect on the animal (for example rat). I mean that if
there is little infect when surgery for collecting blood sample, we
could repeatedly collecting blood from the same animal at different
time points. Or we need arrange a group of animals at different time
points, then just get one-time blood collection from each animal.
I appreciate your kind comments and suggestions very much.
I also have a strong desire to be a volunteer in some Pharmaceutical
companies in this coming winter vocation in order to get more

Do first things first . From your experimental design plans it show
clearly that you will do alot of Blood or plasma samples.
First of all develop a comprehensive Bio-analytical assay for you
compound.Ensure that the analytical method is stability indicating.

I am just wondering whether tritiated 001 or other radio-labeled form
is available for your drug. This would be helpful to assay the
bio.fluids with trace amounts of the drug.

Feng,
I think that you can use fractional factorial experiment design using
replicates in the central point to get your model in your study. With
this procedure you can analise your response according the influence
of various factors like time of drug administration, concentration,
temperature and others.
Talking about the situation number 3, I think that you can collect
your sample in diferrent rats and do the other experiment collecting
all samples in a same rat to compare the results because in the case
of collect sample in different animals you will introduce another
variable in your model.
There are lot of literature disposable and software to work the issue
presented (design of experiment).
