Dear Sir
We are working on the extraction of a drug which is fluorescent and we
use HPLC with fluorescent detector for quantification. We are using a
column (which is safe in the PH range 2 to 10) and we used to extract
our sample through liquid- liquid extraction. Ultimately our sample is
in 0.1 N HCl. We inject about 20 ul. Is it harming column any way. If
yes, then what will be the solution?

HCl is good for a steel column, can you use a different acid? - db]

Hi rahul
Obviously, its not good for your column life. If you use medium 0.1
NHCl, pH will goes below 2.0. You better use waters colum like
Xbridge that are quite stable over wide pH range or change the medium.

The 0.1 N HCl may be an issue. You may want to install a silica
saturation column between the injector and analytical column. The best
approach though would be to try to suspend the analyte in you initial
mobile phase, at the very least you should try to reduce the acidity of
the sample.

Dear rahul,
Tell us about the chemical nature of the drug, I advise you not to use
0.1N Hcl , as it will destroy your column. Intially you will be
getting good results but after about 50 injections your results will
fall down and there will be fluctations. Try to reconstittue the drug
(extracted) in the mobile phase
