Can any one provide the reference for determination of IC50 value and Ki from rat liver microsomes?
It will be great help for studying the DDI.

If you'd want to study DDI I think you should really be using human liver microsomes that are well-characterised.

Dear Nil,
I agree with Prof Pruijn about the relevance and availability of in-vitro systems of human origin (rather than rat) to investigate potential DDI in humans. A similar discussion has taken place on this forum in the last two weeks. A look on the FDA guidance for DDI studies, as well as the PhRMA consensus paper (Bjornsson et al, 2003) might help you, if not already done.
