Dear Khalid,
Thank you very much for your reply =). Since you are quite expert in culturing caco-2 cell line, I just wonder do you have any experience on using the Biocoat HTS caco-2 assay system (for the permeability/absorption study)? If you do, do you think the cell growth will be affected by the special culture media (of the Biocoat HTS caco-2 assay system ) because the caco-2 cell line was cultured initially on the RPMI media before transfering to the insert.

Just as well as a change of fetal calf serum (batch!) may affect
cellular response, the change in culturing you propose will most likely
also influence cell responses. I have no way to assess whether this will
jeopardize your experiment but it may be advisable to use correct
controls (FDA provided some suggestions for benchmarking)

I am sorry to say that i have no experience working with BIOCAT HTS caco-2 culturing system but, from the literature survey, i can say that the HTS system will definitely affect your experimental results in comparison to the traditional 21-day old culture. Secondly, you are transferring the cells from RPMI media to HTS media, so, you may expect change in the morphology of the cells. I would suggest you to first choose the method you want to use, according to your application need, either HTS or traditional.
