Hi,
I have an issue with the nanospheres formulation (PLGA, PVA, Pluronics) for intravenous administration in bolus and infusion in rodents and the drug formulation is available in powder form.
I have tried with phosphate buffer 0.1M and phosphate buffer saline pH7.4 as the vehicle. Formulation is found to be a uniform suspension but the particles is settling down and I am worried about the variation in administration.
Please suggest me a suitable vehicle and the permissible limits of vehicles which can be used for our study.

Hi,
If the suspension leads to nanoparticles sedimentation, the system is
not a colloidal suspension and in addition to pharmacokinetic variation
you may experiment embolism associated with i.v. administration because
of particles aggregation. I think the issue is more about the
nanoparticles size (too big) than the vehicle which in you suspend the
nanoparticles.
