Dear all
We are planning to conduct an exposure study in zebra fish. Coming to sample collection, we planned to collect the samples onto DMPK cards Since the sample volume could be less.
My question is regarding the preparation of calibration curve.
Usually we follow serial dilutions method to prepare the analyte CC concentration in suitable medium like DMSO in such a way the concentrations are sufficient enough to get the ultimation CC concentrations in the matrix (i.e, 1-2ul volume from analyte CC in to the matrix not less than 100ul ). And the processing method would be any of the conventional extraction methods.
As the blood volume in zebra fish is very less, it is quiet challenging to prepare the CC standards.
Can any body suggest me the suitable procedure to prepare CC standards in less volume of matrix.
If iam not clear in any regard, please let me know.

Use a surrogate matrix- from a larger fish or artificially created,
worry about matching matrices when you get into mammals. Express
result as relative to curves and QCs prepared in the surrogate

Dear Vijaykumar
Ideally you have to prepare CC in same matrix as of sample. I understood your limitation is availability of matrix volume! To solve this problem you can cross validate the extraction method with different matrix (Blood from different species), evaluation of recovery will be enough to cross validate the method with two different matrices (zebra fish. Vs matrix X)
If you can develop a method with similar recovery for both the matrix U can use a Calibration curve in validated matrix and easily quantitate your sample

Thanks for your reply.The same idea was with me but in order to cross validate the extraction method , do i need to go for all the CC points or shall i go for limited number(since there is another limitation for DMPK cards).
