This is a very old question asked every year and it was also my own  question 7 years ago. please read a paper by myself (Jiaqiang Cai from  Merck) in next issue of Bioorganic & Medicinal Chemistry Letters on a  cats inhibitor work. we have solved this puzzle for the basic nitrogen  containing compounds.
