Dear Sir/ Ma'am
I am currently doing in vitro CYP enzyme inhibition experiments and i
need help regarding calculation of IC50 values of my test compounds.I am
using GraphPad Prism software version 5 for the same. However, while
using non-linear regression in graphpad prism, it shows some models viz
log inhibitor vs response, log inhibitor vs variable slope, log
inhibitor vs normalized response and log inhibitor vs normalized
response-variable slope. After fitting my data in these models, i am
getting good regression but with different IC50 values for different
models. Kindly guide me regarding the selection of particular model,
what is the criteria of selection. Also, suggest some of the direct ways
of calculating IC50 values. I will be very grateful.
