Dear all,
A test article was given to animals by IV bolus several day before. But
the BA data suggest that the plasma concentration at 5 min < 15 min.
I really feel confused on this. Can anyone give me some explanation for
me? I'll appreciate it if you can give me some documents.

PS: There were many clinical observations within the first 15min, such
as tremors, decreased activities and lethargy. I don't know whether
these symptoms will affect PK behavior or not.

Hello,
What is the IV formulation and how insoluble is your drug?
I had a formulation of trimethoprim where the drug did not remain in
solution once injected into the jugular vein of Holstein calves. The
drug then solubilized and gave a peak 15 to 30 min later.

It can happen probably because of the lag time in transport of drug in
the blood stream.. i hope attached article can help you.. [not attached

But, will there be any lag time in IV route because the Lag time in
pharmacokinetics corresponds to the finite time taken for a drug to
appear in systemic circulation following extravascular administration
i.e not iv . Lag time is a reflection of the processes associated with
the absorption phase such as drug dissolution and/or release from the
delivery system and drug migration to the absorbing surface.
So I doubt there should be some formulation issues only.
Was the study repeated under identical conditions of the previous one?
if so, how the data looks like. There could also be a chance, if the
total drug injected did not enter the systemic circulation, i mean
some amount might be left outside when drawing the needle back and act
as a depot preparation..any further ideas.

Is it possible for the drug to precipitate in lungs? Does it have poor solubility?
