While calculating efflux ratio in MDR1-MDCK cell based permeability screen, is it always
necessary to compare efflux ratio of transfected cell line to that of parent cell line
(non transfected MDCK)?
Could somebody please advise?

Hi Mansi,
Comparing the Efflux ratio in the transfected cell line to the parental cell line gives an
estimate of the true magnitude of P-gp mediated efflux. This is because there is native
P-gp that may be present in the parental cell line and can contribute to drug efflux
(MDCKII is a kidney cell line which can express some P-gp)
Therefore we routinely calculate what is known as the corrected flux ratio (CFR) which is
basically the ratio of efflux ratio in transfected to that in parental cell line. { CFR =
Efflux ratio in transfected cell lines divided by Efflux ratio in parental cell line }
