Dear All
Can any one tell solubility of methotrexate in organic solvent ? DMSO and DMF it is soluble. Literature says that it is soluble in acetone and dichloromethane. But we tried but it could not able to soluble. Any one have best way to solublis will acknowledge.

Methotrexate is class IV drug in short it is having low solubility and low permiability. It is practically soluble in Ethanol,Highly soluble in DMSO and DMF.I would also like to tell you that it is also soluble in Acetone.
Kindly check which form you are using for checking solubility whether crystalline or amorphous.Also note that marketed product of Methotrexate are salt form of Methotrexate.

I tried with normal crystalline form of Methotrexate (MTX) it is soluble in Acetone. Kindly let me know your application area so that I will be able to suggest some better options.

At our lab in BASF India ltd.We tried tried with DMSO,Acetone and DMF.It is soluble in all three.There are some literature in that DMSO is used for preparation of nanoparticals.I will suggest to use acetone as it will easily get evaporated. Also try to use Albumin for preparation of nanoparticals.
