Gary,
WNL classic did not offer a 3-compartment extravascular model as part of the
pharmacokinetic model library since they are often over-parameterized if you only have
concentrations measured in one compartment (e.g. systemic circulation).
I don't have a WNL classic 'ascii' model with 3 compartments and oral input to hand
but if you take your free upgrade to Phoenix WinNonlin you will have access to the new
Phoenix model object where a 3 compartmental Extravascular model is built-in and it's
easy using the graphical or textual editors to change the default to one with e.g. a deep
depot for the 3rd compartment.
Furthermore with the easy implementation of NLME functions you are more likely to be
able to fit your data with more confidence using population methodology if you wish.
