Dear Colleagues.
If drug have low water solubility and lower absolute bioavailability (F < 0.3),
then is it possible to place it BCS class IV?

I tend to hesitate using bioavailability as a BCS criteria since it is as
much a function of formulation as an inherent physical property of the
compound. Case in point is itraconazole, which is listed as a Class II
compound. Given the wrong formulation, its absolute bioavailability is very
low and, as a compound of very low solubility, would be, I think, wrongly
classified as Class IV.

BCS classification is based on the compound solubility and permeability =
so we
should have solubility and permeability data with us to classify the =
compound.
This compound shows very low BA so we can say low permeable but there =
are number
of other factors responsible for low BA.=20

If drug dose solubility ratio is less than 250 mL in the entire pH range =
of 1 to
7.5 it is considered as Highly soluble and absolute bioavailability is =
greater
than 85% is considered to be highly permeable

Metabolism.
Then, we can not to affirm 100% that drugs with low water solubility and =
lower
bioavailability are class IV.=20
