Dear All,
I am working on Solid Lipid Nanoparticles (SLNs) which is a relatively new carrier system
for drug. It undergoes selective Lymphatic absorption and protects the drug from high
first pass metabolism. I would like to know if anybody is working on lymphatic uptake
mechanism(s)?
My objective is to develop a model showing that drug-carrier complex is undergoing
lymphatic uptake. Therefore what studies are to be done to support the statement.
Your expert comments would be highly appreciable

Dear Rahul,
I am doing some work in that area too, and there is little in the literature
done before. The most articles you can find is about modelling of antibodys,
that go through the lymphatic system as well.
For your modelling exercise it would be very usefull to have drug concentrations
of plasma and lymph at the same times, if possible. And if you want to see how
the drug releases from the complex in-vivo there are some imaging tecniques with
isotopes marking the particle as well as the drug, although I heard it's quite
expensive.

Many thanks for your expert comment on Lymphatic uptake.
My objective is to see the concentration of drug in Lymphatic system when
administered though oral route.
I tried lymphatic cannulation both thoracic and mysentric but found it very
difficult.
Is there any paper or report on cannulation technique or any other useful
suggestions which may improve my technique of cannulation.

Dear Rahul,
Lymphatic cannulation surgery is li'l bit difficult. as lymph ducts will rupture
as you insert the cannulae.
But for proper visibility you can do one thing, first administer approx 2-3 ml
edible oil to rat orally, after 20-30 min anaesthetise the animal and c, you 'll
have very clear view of lymph ducts.
Another one is After anaesthesia administer evan's blue or congo red dye
directly to tesitcle within few minutes it will absorb to lymph due to its high
molecular weight and u'll have clear idea. After practicing on these animals may
it will be easy with normal animals.Try to do surgery with soft silicon tubing
having very small o.d. (less than 0.6 mm) and fix the cannula to internal
abdominal wall with some sutures will avoid dislocation.
It might help you somewhat !!
