Hello group,
In PBPK modeling for predicting in vivo CL, for few NCEs when I calculated intrinsic
clearance from Ln peak area ratio Vs time graph, NCEs are falling under the low clearance
category but when the same calculation is done with % remaining Vs time graph, intrinsic
clearance value is falling under high clearance category. i.e. contradicting conclusions.
Further, when above intrinsic values were used to predict the in vivo clearance by
well-stirred model/parallel tube model the contradiction continued. Which is the method
one should consider for accurate prediction? Let me know your thoughts on this.
