dear all,
does anyone have an experience in estimating pk of drugs in breastmilk, the guideline of research
when conducted in human, and i would like to have the most cited references. thank you in advanced.

What exactly are you asking for - estimate of pKa in breast milk, or estimate of PharmacoKinetics
(lactational transfer) in breast milk?
Best wishes.

We had done sparse sampling study design to assess the transfer of
tramadol and its O-desmethyl metabolite into transitional breast milk,
as well as model it. Professor Kenneth Ilett from the University of
Western Australia had done a lot of work in this area. I have listed
the two references that I mentioned below.
Ilett KF, Paech MJ, Page-Sharp M, Sy SKB, Kristensen JH, Goy R, Chua
S, Christmas T, Scott KL. Use of a sparse sampling study design to
assess transfer of tramadol and its O-desmethyl metabolite into
transitional breast milk. Br J Clin Pharmacol 2008;65:661-6.

pharmacokinetic modeling of tramadol and its O-desmethyl metabolite in
plasma and breast milk. Eur J Clin Pharmacol 2011; 67:899-908.

Thomas W. Hale runs infantrisk.com would be a very good go-to for this information on many drugs.
Often the studies aren't great (limited subjects, limited time points) but better than nothing. Keep
in mind that the amount found in milk at the beginning of a session and the amount at the end of the
session may vary due to fat content and, I would assume, time to equilibrium.

Thank you for the information, actually i want to learn about the absolute dosage an infant will
receive from his/her mother's milk. bu I am not sure about the analysis methods or sampling time.
very best regards,
