Dear all,
Can anyone recommend formulations that are routinely used for dog PK (iv/po). We hardly carry out
these studies and would appreciate some help.

Hello,
This question is a bit vague. What first needs to be asked is what is the question you are trying to
answer? Are you doing a bioavailability study where you need both iv and po routes? It sounds like
that may be that case. Is this a drug that is already developed for these routes or do you need to
have it compounded?
And in terms of formulation for PO what is the intended use for this drug? Is it a liquid
suspension, oral tablet, time-released tablet???
Hope these questions will help guide you in determining what you are actually trying to ask. The
group will be able to assist you better if you could clarify your question.

Hello,
Sorry for intervention.
Question arises whether both route of administration is needed for PK studies and for regulatory
submission?
if the oral study is available, would it suffice if the same parental drug is prepared for IM and
IV administration and still studies need to be shown ?(ex diclofenac/ aceclofenac?

Is this in development or discovery? In discovery, I have seen almost any solvent used. Some
solvents can be very toxic and that toxicity is often discounted in discovery. In development there
is much more consideration given to limit the toxicity to the API alone and for obvious reasons. So
the question to you is are you in discovery or develoment?

Dear Dr. Florence,
It depends on the nature of molecule you are working upon. whether it is water soluble or not. If
molecule is water insoluble, may use vitamin based surfactants to solubilize the compound. It would
facilitate the IV/PO administration of formulation for animal studies.
