Dear all,
Why the incubation time is 4h in PAMPA assay? Has the equilibrium been reached at 4h? If so, we
cannot differentiate those cpds reaching equilibrium less than 4h and Pe of these cpds could be
underestimated based on related formula.
However, if the purpose is not to reach equilibrium at all at 4h, why don't we shorten the
incubation time to 2h?

[My understanding is that PAMPA is a transport assay and presumably the 4 hr is design to ensure
sufficient time for measurable transport across the 'membrane' - db]

It is indeed a good question. In fact we carry out incubation for 19 hr.
I do also have Q regarding incubation temperature. Is it okay to carry out at room temperature or
one need to incubate at 37 C?
So please elaborate your views about PAMPA on this aspect also.
