PharmPK Discussion - Metabolism study

  I want to perform metabolism studies for 2 compounds ( curcumin and Dimethoxy curcumin) in order to
  compare between their intrinsic clearence.
  
  I have done the experiment several times and in each time the intrinsic clearence of
  dimethoxycurcumin is higher than curcumin which is opposite to literature.
  
  I found that the wrong in my procedure that i was subjecting them both only to Phase I metabolism
  which is not the true case invivo; so i think i have to include Phase II metabolism by the use of (
  Uridine 5'-diphosphoglucuronic Acid Trisodium) which is important in gluronidation but i don't know
  whether i have to include phase I and II together in the same step and if so, what will be the
  procedure.
