Dear all,
I am trying to model plasma concentration profile of 10 patients after oral administration of 200 mg
of progesterone capsules (Prometrium) using Winnonlin. Does anyone know whether it follows one or
two compartment model? Further, I want to use the parameters obtained for simulating plasma
concentration of another oral formulation of progesterone.

Dear Pinak,
You can confirm the model (1CM or 2CM) by checking firstly in-built models/writing differential
equations in WinNonlin and comparing with the observed data.
You can firstly check the fit by visual inspection and then by checking he diagnostics (AIC, SBC,
WSSR) (look for model that has least value of each diagnostic at same weighing scheme) and plots of
" Predicted Y vs Observed Y"
Before plugging in the data in WinNonlin you can also check the semi-log plot of plasma vs time. The
mono-exponential decline of the conc. vs time curve will indicate 1-CM and a bi-exponential decline
will indicate 2-CM.
