Dear fellow pharmpk readers,
I am trying to determine the erythrocyte binding constant (kd) and bmax of a drug, based on
information in the FDA clinical pharmacology and biopharmaceutics review. I have extracted
quantitative data from a graph showing relative distribution of a drug in erythrocytes and plasma
versus whole blood concentrations.
For contruction of a scatchard plot, I plot the bound:free ratio on the Y-axis, but as the X-axis
should contain the specific binding, I am confused what I should use here. Should I put the whole
blood concentration there, or should I correct for hematocrit (which is unknown)?
