PharmPK Discussion - Relationship between ka and Peff

  Dear All
  We performed the in-situ studies for intestine of developed nano
  formulations and calculated ka using below formula.
  
  lnXr = lnX0 - ka x t
  
  t(1/2) = 0.693 x ka
  
  For pure drug there is increase in ka and followed by saturation at 1.5hr
  and for developed formulations ka was increased till 3.5 hrs and followed
  by saturation.
  The query was raised what is Peff from in-situ study. Can any one put
  light on this aspect or any suggestions how to justify.
  
  Can i say the absorption rate enhance with saturation times indicates
  increase in permeability?
