PharmPK - Discussions about Pharmacokinetics
Pharmacodynamics and related topics
I am deciding to perform a clinical project in evaluating the netilmicin
pharmacokinetics parameters on Chinese neonates. Does anyone have the data
or know where to find any relevent information regarding the design of the
study and any relevant references that I can refer to?

You may contact Dr. Roger Jelliffe, he has done excellent work in this
area. His coputer program will be very helpful. I have attached his web

Dear Harry:
Netilmicin (N) is like gentamicin and tobramycin with respect to its PK
properties.
Therefore renal function will be an important factor. The recommended dose
(Goodman and Gilman, 9th Edition, page 1116) is 4 to 6.5 mg/kg per day in 2
divided doses. by infusion; t1/2 of N can be as long as 37 hrs in neonates
(ibid, page 1765) Briefly, the following (preliminary) protocol is
recommended:
Administer the drug by infusion (3 mg/kg) over 1 hr, draw blood samples at
0.25,0.5, 2, 4, 6, 8, 12, 24, 36, 48 (post infusion) in 10-12 neonates
whose renal function (creatinine clearance) is known.
Determine plasma concentrations of the drug.
From the post-infusion data, you should be able to determine the PK
parameters (such as t1/2, Vd, clearance).
Correlation analysis between PK parameters and renal function should be
useful in preparing nomogram for drug dosing.
Please contact me directly if I can be of further assistance.
Sincerely,
Sri
