Hi,
We are trying to establish automatic blood collection from venous
cannulated rats.
I am familiar with the microdialysis constant sampling approach, and we can
use syringe withdraw pump or peristaltic pump for collection over a single
interval, or by adding in switching valves and a fraction collector, do
continuous withdraw and collect continuous sampling-fixed time fractions.
What we would like to do is automatically collect fixed volume samples at
various times. In between samples we want to wash the loop and cannula
clean.
Does anyone knows how to do this or is aware of a reference where fixed
volume variable time automatic sampling is described?

You can backflush with heparin, it presents risk, too much and the animal
may bleed out, also the following samples may become diluted with the
heparin wash, a correction can be made by quantifying the heparin with your
assay, secondly the PK's may be effected if your study involves the
coagulation pathway.
