Dear All:
One compound is currently under clinical trial. This compound undergoes
hepatic biotransformation to its corresponding glucuronide by uridine
diphosphate glucuronosyltransferase (UGTs).
One study was done using 72h infusion and reported enterohepatic
recirculation (a post-infusional increases in plasma concentrations,
generally occured between 3 and 24 h after the end of infusion) in 22 of 78
patients. However, no such post-infusisn increase was observed using 1-h IV
bolus in 50 patients.
The t1/2 for 72-h infusion is 11hr and 4 hr for 1h bolus.
Question: why no post-infusion peaks were observed in 1-h IV bolus?
Difference of t1/2 could be due to enterohepatic recirculation,
any other reasons?
