Hi - I am not sure what you mean by Therapeutic Index. We did a study
of ibuprofen in young and elderley (re. Kings College Hospital with
permission) but didn't dose by infusion. However I have results for
single bolus dose of 400mg ibuprofen which may help. Mean AUC(0-inf)
for the unbound R- and S-enantiomers in healthy young subjects were 147
and 356 ng.h/ml. The results for total R- and S- enantiomers were 63.6
and 82.8 ug.h/ml respectively. The corresponding values for Cmax were
37.8, 70.2 ng/ml and 16.0 and 16.6 ug/ml. As you can see the S(-)
enantiomer is predominant and is the pharmacologically active isomer.
You will have to take the interconversion of S- to R- into account as
this is causes loss of therapeutic activity following dosing.
