Hi,
I am trying to calculate absorption PK for a drug (paracetamol) in healthy adult volunteers.
Initial though was to use the method of residual to calculate, but I don't have IV data for this
study.
Could someone tell me how I can go about doing this. I was thinking of using data from previous
publications (iv data) is this a right approch?

Hello Radhakisham,
If you have sufficient number of conc.-time sample data points (minimally, about 8-10) the method of
residuals (curve stripping) can define the kinetic rate and intercept values for up to three
distribution phases (compartments) such as absorption, distribution, and terminal elimination. Or
serve as seed values for iterative analyses (e.g., curve fitting and compartmental modeling).

Radhakrishnan:
You will need to use deconvolution to do this you will need IV and PO data from the same subject,
preferably at the same time! Please look up the "semisimultaneous method" and deconvolution.
