Hello all,
Food effect from a high-fat meal typically delays stomach empting and Tmax by 1-2 hrs for most
drugs, but I am wondering if there are published papers (reviews, individual compounds, M&S) looking
at food effect on Tmax for drugs that undergo EHC. Intuitively, food intake could trigger EHC and
should further prolong Tmax. What is your experience?
Thanks,
Jack

Hello all
you can look for clopidogrel and their metabolite as example
thanks

I don't think clopidogrel is a good example as the drug itself does not undergo EHC but thanks for
mentioning it. Diclofenac or rofecoxib are more interesting molecules (for this ADME feature), but
since most food effect studies are conducted with a single dose, a significant influence on Tmax by
EHC perhaps is not to be expected. In my case, the food study was conducted at PK steady-state. I
have other evidence to support EHC for the drug, but not sure if I should relate a food induced
4-hour Tmax increase to it. Tx.
