Dear All,
I am using MDCK-MDRI cell monolayer to do the screening of pgp
substrates, I will test lots of compounds in the same batch, if I use
standard curve to detect compound concentration , that should be a lot
of work, However I want to use the intensity(peak area)(LCMS) to
calculate the Papp directly so that to avoid preparing lots of compound
standards.
The question is , how can I calculate the Papp, using the formulation
Papp=dQ/dt*A*C
the unit can not be cm/s...

Papp in (cm/s)=(dQ(moles of drug in either 750uL for A->B or 250uL for
B->A))/dt(sec)*0.33(cm2)*C0 (moles/cm3))
so here is how I do it in costar 24 well traswells where the top
compartment is 250uL and the bottom compartment is 750uL.
The one that got me was that last unit for C initial moles/cm3 , that is
to say moles/mL
