I hope this isn't too elementary, but I can't find anything in my
Applied PK text, how can you calculate bioavailability without an IV
comparator?

you need IV PK to estimate the absolute bioavailability, or you can
estimate the relative bioavailability by comparing to an oral solution or to
a known dosage form.
