i would like to know something about the metabolism of troglitazone(like
which enzymes are responsible for the metabolities) and also if more is
known about the metabolite which may cause any/some hepatic effects.
please give references if possible.

Dear Kartik,
There is a review article on troglitazone in the March 1998 edition of
Annals of Pharmacotherapy by Johnson, M.D. et al page 337-348. In this
article they cite one published article on the pharmacokinetics of
troglitazone in humans (Izumi et al J.Pharmacol Exp Ther 1996;277:1630-41).
Otherwise the pharmacokinetic data thay they use was obtained from the files
of the manufacturer (Parke-Davis).
They report that troglitazone is metabolized by the CYP450 isoenzymes
to some extent, and may interact with drugs which are metabolized by these
enzymes.
It is metabolized to a sulfate and quinone of the parent compound, which is
an alpha-tocopherol substituted thiazodolidinedione.
