Hello All-
I am interested in conducting small study of acetaminophen pharmacokinetics
in my undergraduate students in order for them to better understand
(hopefully) pharmacokinetic principles. I need to use salivary
concentrations (non-invasive) and was wondering if any of you know of a
commercial (or even homemade) sample collector, like an oral sponge. I seem
to recall something being commercially available, but do not remember much
other than I think one exists. Any help would be greatly appreciated.

When I was at NASA we purchased our salivary sample collectors from
Sarstedt. I don't know their number or address, but I am sure somewhere
around UT someone has it.

In a current study of Pk-Pd relationships of aminocetophen (which we call
paracetamol) we are simply collecting saliva in universal specimen tubes
about 1 inch in diameter so that volunteers can easily use them. Most
specimen tubes are rather too narrow for convenience. Samples have to
spun down to precipitate debris, but that is probably no more messy than
retrieving the drug from a sponge.

Lane,
NASA pharmacology uses Salivettes for the collection of saliva from
astronauts during flight. They have also modified these devices to
incorporate a preservative to prolong sample integrity during storage
at ambient temperatures. The device consists of a cylindrical
polypropylene tube which contains a gauze roll about the size of a
cigarette filter. The subject places the gauze roll in the mouth for
several minutes until it is saturated with saliva and then it is
returned to the tube. The device is placed into a centrifuge and a
short, low G spin extrudes the saliva into a lower chamber from which
it can be harvested for analysis.

Dear Lane,
Try a SALIVETTE, a citric acid impregnated spongelike thing to chew on
which stimulates salivary and collects it. When you have chewed on it
you can easili centrifugate it and collect the saliva.

For saliva collections it is a common practice to take a small piece of
parafilm sheet commonly available in any pharmaceutical/clinical laboratory
chew this or simply place it below the tongue. This gives enough stimulation
to secrete saliva. This is a common practice in many labs performing
antipyrene metabolism. Caution must be exercised if the analytical method
for your assay in a Mass Spectrometric method. In mass spec methods
parafilm presents many interfering peaks.

Dear colleagues,
With respect to saliva sampling: please note that the method for
sampling saliva (e.g. intensive chewing, citric acid) may influence
the drug concentration. I remember that I have read about that topic
many years ago.
